Effect of various sample-handling conditions on radioassay results.

چکیده
Samples brought to the laboratory for routine radioassay often have been under suboptimal conditions. We compared the effect of some sample-handling conditions on the values for T3, T3 resin uptake (T3RU), T4, thyrotropin, cortisol, and prolactin in serum as determined by radioassays. Blood was collected in three separateplain red-top Vacutainer Tubes from each of six subjects. Serum was separated from one of the three tubes, divided into portions, and frozen within 1 h of collection. Another tube from each patient was allowed to sit at room temperature for 7 h and then the serum was separated, apportioned, and frozen. The third tube from each patient was allowed to sit at room temperature for 7 h, then left overnight in the refrigerator and the serum separated and frozen after a further 17 h. All assays were done with commercially available kits: T4, T3RIA, and thyrotropin kits from Beckman Instruments Inc., Fullerton, CA 92634; T3RU kits from E.R. Squibb and Sons Inc., Princeton, NJ 08540; cortisol kits from Clinical Assays, Cambridge, MA 02139; and prolactin kits from Abbott Laboratories, Diagnostics Division, North Chicago, IL 60064. We could detect no significant detrimental effect of these various samplehandling conditions on the results of routine radioassays.

Evaluating the Effect of Various Parameters of Protective Spur Dike on Scour Depth Reduction using Group Method of Data Handling (GMDH) and Gene Expression Programming (GEP)
Spur dikes are one of the common methods to protect rivers against erosion. Scouring around the spur dike is an important factor that can disorder the structural performance. Using protective spur dike is proper technique reduce the scour amount. In this research, the GMDH and GEP model used in order to evaluate and estimate the effect of various parameters of protective spur dike o...
متن کاملClinical and laboratory observations on serum folate-binding protein.
We studied the effect of serum folate-binding protein (FBP) on folate radioassays and the relationship of the serum level of unsaturated FBP to the serum folate level in various clinical states. Our modification of a heat-extracted radioassay was compared to a whole serum radioassay. Our results confirmed the existence of elevated serum levels of unsaturated FBP in some normal subjects, in some...
متن کاملThe Development of a Passive False Twister Mechanism in Handling Low Strength Cotton Slivers on High Draft Spinning Machine
A passive false twist unit (spiral) has been developed to assist with the handling lowstrength slivers on a high speed-spinning machine with a high-speed feed. In the first trial, a falsetwist simulator device was constructed to determine whether the passive false twist unit can be usedon high speed feeding with different can distance from the feeding device. In the second trial, theeffects of ...
متن کاملRadioassay for cobalamin (vitamin B12) requiring no pretreatment of serum.
We describe a radioassay for cobalamin (vitamin B12) in human serum or plasma that requires no boiling or other pretreatment of the sample. Normal chicken serum covalently coupled to magnetizable particles is used as the binding agent. The assay is performed at pH 12.9, at which pH all cobalamin in human serum is released from its binding proteins, whereas the binding agent maintains a high aff...
متن کاملAnti-dsDNA antibody assay: high specificity and sensitivity with a filtration radioassay in comparison to low specificity with the standard ELISA.
OBJECTIVE To evaluate whether a new fluid-phase filtration radioassay possesses both high sensitivity and specificity compared with the currently used ELISA and Farr assays. METHODS Sequential sera (25 samples) from 9 patients with systemic lupus erythematosus (SLE), sera from 20 patients with SLE possessing anti-dsDNA antibodies by the Crithidia assay, 75 patients with rheumatoid arthritis p...
